Brian Dorsey here. We spoke on the phone about submitting a MiSeq library. FYI, I've run 14 cycles of PCR to attach flow cell adapters and get the concentration to 14 ng/ul in ca. 25ul. Please see the conditions for the run below.
Please size select the library to a mean insert size of 250 bp. I would like there to be some overlap to compensate for the lower quality calls at the 3' end.
Please sequence 150 bp paired-end reads using the Micro mode on the MiSeq.
There are inline barcodes after the 5' sequencing primer but only one (common) index sequence for all individuals in the library.
